Overview

Methadone + acetaminophen is a combination of two analgesic agents used together for the management of pain. Methadone is a synthetic opioid analgesic that acts primarily as a full agonist at the µ-opioid receptor, with additional activity as an antagonist at the N-methyl-D-aspartate (NMDA) receptor and inhibitor of serotonin and norepinephrine reuptake. It is indicated for severe pain not responsive to alternative treatments and for opioid dependence therapy[3][6][10]. Acetaminophen (paracetamol) is a non-opioid analgesic and antipyretic agent, commonly used to treat mild to moderate pain and fever by inhibiting prostaglandin synthesis in the central nervous system[5]. The combination may be considered when both opioid-level pain control and non-opioid adjunctive therapy are needed. There are no highly clinically significant interactions reported between methadone and acetaminophen, but clinical monitoring remains prudent[1].
