This multi-agent chemotherapy regimen was developed by Memorial Sloan Kettering Cancer Center (MSKCC) for the treatment of newly diagnosed primary central nervous system lymphoma (PCNSL). The protocol consists of an intensive induction phase using high-dose methotrexate and cytarabine (Ara-C), which are selected for their ability to penetrate the blood-brain barrier. This is followed by a consolidation phase involving high-dose BEAM chemotherapy—comprising carmustine, etoposide, cytarabine, and melphalan—and autologous peripheral blood stem cell transplantation (ASCT). The regimen aims to achieve deep remission through antimetabolite-mediated inhibition of DNA synthesis during induction, followed by myeloablative conditioning to eliminate residual disease in the CNS and systemic compartments.
