Overview

This is a combination chemotherapy regimen consisting of three antineoplastic agents—methotrexate, mitoxantrone, and vincristine. Each drug has a distinct mechanism of action targeting cancer cell proliferation: - Methotrexate is an antimetabolite that inhibits dihydrofolate reductase, blocking DNA synthesis and cell replication. - Mitoxantrone is an anthracenedione that intercalates into DNA and inhibits topoisomerase II, leading to DNA strand breaks and apoptosis. - Vincristine is a vinca alkaloid that binds to tubulin, inhibiting microtubule formation during mitosis and causing cell cycle arrest at metaphase. The combination has been studied as first-line therapy for advanced breast cancer (MIMO regimen), showing objective response rates around 42% with manageable toxicity profiles. It has also been used in other malignancies in various combinations[2][8].
