Drug intelligence / Profile preview
Methotrexate + olokizumab is a combination therapy used for the treatment of moderate to severe rheumatoid arthritis, particularly in patients who have had an inadequate response to methotrexate alone. Methotrexate is a folic acid antagonist that inhibits dihydrofolate reductase, leading to immunosuppressive and anti-inflammatory effects. Olokizumab is a humanized IgG4 monoclonal antibody that directly binds to interleukin-6 (IL-6) at site III, thereby inhibiting IL-6-mediated proinflammatory signaling and suppressing the JAK-STAT pathway. This combination has demonstrated efficacy in reducing disease activity and improving patient-reported outcomes in multiple phase 3 clinical trials, with safety and tolerability maintained over long-term use[1][2][3][4][5][6].
