Overview

This is a combination chemotherapy regimen consisting of three antineoplastic agents—methotrexate, peplomycin, and vindesine. Methotrexate is an antimetabolite and antifolate agent that inhibits dihydrofolate reductase, thereby blocking DNA synthesis and cell replication. Peplomycin is a glycopeptide antibiotic related to bleomycin; it induces DNA strand breaks through the generation of free radicals in the presence of iron. Vindesine is a vinca alkaloid that disrupts microtubule formation by binding to tubulin, inhibiting mitosis. This combination has been used experimentally for intraarterial chemotherapy in glioblastoma and potentially other malignancies[2][3]. The regimen aims to exploit different mechanisms of action for synergistic cytotoxicity against cancer cells.
