Methotrexate + vinblastine + doxorubicin + cisplatin + panitumumab is an investigational combination regimen evaluated for the treatment of advanced urothelial carcinoma. This regimen combines the dose-dense or intensified MVAC (dd-MVAC) chemotherapy backbone—comprising methotrexate (an antimetabolite), vinblastine (a microtubule inhibitor), doxorubicin (an anthracycline), and cisplatin (a platinum-based alkylating agent)—with panitumumab, a recombinant human IgG2 monoclonal antibody that binds to the epidermal growth factor receptor (EGFR). Developed and studied by UNICANCER in the randomized Phase II trial NCT02818725, the combination was specifically tested in patients with KRAS and HRAS wild-type tumors. Clinical findings indicated that the addition of panitumumab to intensified MVAC did not provide a survival benefit and resulted in higher toxicity compared to chemotherapy alone.
