Overview

Methylphenidate + guanfacine is a combination therapy used primarily in the treatment of attention-deficit/hyperactivity disorder (ADHD), especially in children who have an inadequate response to monotherapy. Methylphenidate is a central nervous system stimulant that inhibits the reuptake of norepinephrine and dopamine, increasing their concentrations in the synaptic cleft. Guanfacine is an alpha-2A adrenergic receptor agonist that acts on prefrontal cortical receptors to enhance executive function and reduce hyperactivity and impulsivity. The combination may provide greater improvements in ADHD symptoms than either drug alone, particularly for visuo-attentional deficits and working memory performance[3][4][8]. However, combined use can affect cardiac rhythm and requires monitoring for potential side effects such as irregular heartbeat or changes in blood pressure[1].
