This combination therapy consists of methylprednisolone, a synthetic glucocorticoid, and mycophenolate mofetil, an immunosuppressive antimetabolite. It is being investigated by researcher Xiaoli Fan at West China Hospital for the treatment of primary biliary cholangitis-autoimmune hepatitis (PBC-AIH) overlap syndrome. Methylprednisolone acts as an anti-inflammatory agent by binding to glucocorticoid receptors, while mycophenolate mofetil is a prodrug of mycophenolic acid that inhibits inosine monophosphate dehydrogenase (IMPDH), thereby blocking the de novo synthesis of guanosine nucleotides and selectively inhibiting T and B lymphocyte proliferation. The combination aims to induce remission in treatment-naive patients by targeting the dual autoimmune and inflammatory pathways of the overlap syndrome.
