Overview

Metoprolol + amiodarone is a combination of two pharmaceutical agents used primarily in the management of cardiac arrhythmias and related cardiovascular conditions. Metoprolol is a selective β1-adrenoceptor (beta-1 adrenergic receptor) blocker that reduces heart rate, myocardial contractility, and blood pressure by antagonizing sympathetic stimulation of the heart. Amiodarone is a class III antiarrhythmic agent with complex electrophysiological effects, including prolongation of the action potential duration and refractory period in cardiac tissue; it blocks potassium channels as well as sodium and calcium channels, and exerts noncompetitive antiadrenergic effects. The combination has been studied for its efficacy in improving cardiac function after myocardial infarction (MI), reducing arrhythmia recurrence (notably atrial fibrillation), lowering inflammatory markers, and enhancing immune function. While both drugs can independently cause bradycardia or conduction disturbances, clinical studies suggest that their combined use does not significantly increase these risks compared to monotherapy when appropriately monitored[1][3][5][6]. The combination may be more effective than either drug alone for certain indications such as maintaining sinus rhythm after MI or surgery[5][7].
