This investigational fixed-dose combination (FDC) therapy, often referred to as the POLY-HF polypill, is being developed by researchers at the University of Texas Southwestern Medical Center specifically for the treatment of heart failure with reduced ejection fraction (HFrEF). The formulation combines three guideline-directed medical therapies (GDMT) into a single daily capsule: metoprolol succinate (a beta-1 selective adrenergic receptor antagonist), spironolactone (a mineralocorticoid receptor antagonist), and empagliflozin (a sodium-glucose cotransporter 2 inhibitor). By simplifying the treatment regimen, the polypill aims to improve medication adherence and clinical outcomes, particularly in underserved and racially diverse patient populations. Results from the Phase II POLY-HF trial demonstrated significant improvements in left ventricular ejection fraction and a reduction in heart failure-related hospitalizations and emergency visits compared to usual care.
