mezagitamab + bortezomib + lenalidomide + dexamethasone is an investigational combination regimen consisting of a monoclonal antibody (mezagitamab), a proteasome inhibitor (bortezomib), an immunomodulatory agent (lenalidomide), and a corticosteroid (dexamethasone). Mezagitamab is a fully human IgG1 monoclonal antibody targeting CD38, designed to deplete plasma cells, plasmablasts, and natural killer cells, with associated immunomodulatory effects[3][5]. Bortezomib inhibits the 26S proteasome, disrupting protein degradation in malignant cells and inducing apoptosis[1][6]. Lenalidomide exerts immunomodulatory, anti-angiogenic, and direct antineoplastic effects, including enhancing cytotoxic T cell and NK cell activity and inhibiting tumor-supporting blood vessel formation[4][6]. Dexamethasone, a synthetic corticosteroid, is used to manage inflammation and increase the antitumor effects of the other agents[4]. The combination aims to improve outcomes in relapsed/refractory multiple myeloma (RRMM) by synergistically targeting malignant plasma cells through complementary mechanisms, with mezagitamab providing additional immunomodulatory activity[5].
