Overview
**Mezigdomide + daratumumab + dexamethasone** is a combination therapy under clinical investigation for **relapsed/refractory multiple myeloma**. - **Mezigdomide** is an oral *cereblon E3 ligase modulator (CELMoD)* that induces degradation of transcription factors Ikaros and Aiolos, resulting in apoptosis of myeloma cells and immunomodulation. - **Daratumumab** is an *anti-CD38 monoclonal antibody* that targets and leads to the death of CD38-expressing myeloma cells via immune-mediated mechanisms. - **Dexamethasone** is a *synthetic glucocorticoid*, acting as an immunosuppressant and anti-inflammatory agent. The combination is designed to enhance tumoricidal and immune-stimulatory effects against multiple myeloma, especially in patients previously exposed to other myeloma therapies. The regimen is not yet approved and is currently being evaluated in phase 1/2 clinical trials, such as CC-92480-MM-002, for its efficacy and safety[1][4][5][6].
