mFOLFOX6 + durvalumab is a combination regimen used in oncology, consisting of the chemotherapeutic regimen mFOLFOX6 and the immunotherapy durvalumab. mFOLFOX6 is a variation of the FOLFOX chemotherapy regimen, comprised of oxaliplatin, leucovorin (folinic acid), and fluorouracil (5-FU), used primarily for colorectal and gastrointestinal cancers[3][4][6]. Durvalumab is a human IgG1κ monoclonal antibody that inhibits programmed death-ligand 1 (PD-L1), thus enhancing antitumor immune responses. The combination is being studied in clinical trials for synergistic efficacy, with mFOLFOX6 targeting proliferating tumor cells via DNA crosslinking and antimetabolite action, and durvalumab blocking immune suppression by PD-L1 to allow T-cell mediated killing of tumor cells.
