This is a combination regimen consisting of the chemotherapy protocol mFOLFOX6, the immune checkpoint inhibitor durvalumab, and the investigational monoclonal antibody monalizumab. **mFOLFOX6** is a modified version of the FOLFOX chemotherapy regimen and includes oxaliplatin, leucovorin (folinic acid), and 5-fluorouracil. It works by damaging DNA and inhibiting cell division in rapidly dividing cancer cells[1][2][3][4]. **Durvalumab** is a human monoclonal antibody that blocks Programmed death-ligand 1 (PD-L1), preventing it from binding to Programmed cell death protein 1 (PD-1) on T cells, thereby enhancing anti-tumor immune responses. It is developed by AstraZeneca. **Monalizumab** is an investigational humanized IgG4 monoclonal antibody targeting NKG2A receptor on natural killer cells and some T cells; it blocks inhibitory signaling to enhance anti-tumor immunity. Monalizumab was developed by Innate Pharma in collaboration with AstraZeneca.
