mFOLFOX6 + panitumumab is a combination regimen used primarily as first-line therapy for patients with metastatic colorectal cancer (mCRC) who have wild-type RAS genes, as well as for small bowel or appendiceal cancers. The regimen consists of the chemotherapy protocol mFOLFOX6—which includes oxaliplatin, 5-fluorouracil (5-FU), and leucovorin—combined with panitumumab, a monoclonal antibody targeting the epidermal growth factor receptor (EGFR). Panitumumab binds specifically to EGFR on both normal and tumor cells, competitively inhibiting ligand binding and preventing receptor autophosphorylation. This results in inhibition of cell growth, induction of apoptosis, decreased production of proinflammatory cytokines and vascular growth factors, and internalization of EGFR. The combination has demonstrated improved overall survival and response rates in patients with RAS wild-type tumors compared to chemotherapy alone[1][4][8].
