mFOLFOX6 and CapeOx are standard-of-care adjuvant chemotherapy regimens for colorectal cancer. mFOLFOX6 consists of oxaliplatin, leucovorin (folinic acid), and 5-fluorouracil (5-FU), typically administered via intravenous infusion. CapeOx (also known as XELOX) consists of oral capecitabine and intravenous oxaliplatin. Both regimens aim to eliminate residual cancer cells post-surgery by inhibiting DNA synthesis and repair. Oxaliplatin acts as a platinum-based DNA cross-linking agent, while 5-FU and its prodrug capecitabine inhibit thymidylate synthase. Leucovorin is used in mFOLFOX6 to enhance the binding of 5-FU to its target. In the context of the provided trial, these regimens serve as the control arm for comparison against the more intensive mFOLFOXIRI regimen in patients with stage IIIB or IIIC colorectal cancer.
