mFOLFOXIRI + cetuximab is a combination chemotherapy regimen used primarily for the treatment of metastatic colorectal cancer, particularly in patients with RAS and BRAF wild-type tumors. The regimen consists of a modified FOLFOXIRI backbone—oxaliplatin (85 mg/m²), irinotecan (165 mg/m²), folinic acid (400 mg/m²), and 5-fluorouracil (2,800 mg/m² as a 46-hour continuous infusion)—administered every two weeks. Cetuximab, an epidermal growth factor receptor (EGFR) inhibitor monoclonal antibody, is added at 500 mg/m² IV every two weeks. The mechanism of action involves cytotoxic effects from the chemotherapeutic agents combined with targeted inhibition of EGFR signaling by cetuximab to suppress tumor cell proliferation and survival[4][5]. This combination aims to increase response rates and resectability in metastatic colorectal cancer.
