MG005 + sorafenib is a combination therapy consisting of **MG005** (also called **GW5074**) and **sorafenib** designed for the treatment of advanced or metastatic solid tumors. **MG005** is a small molecule inhibitor of CRAF (Raf-1 kinase) that induces cancer cell necroptosis via mitochondrial dysfunction. **Sorafenib** is a multikinase inhibitor with inhibitory activity against multiple protein kinases, including CRAF, BRAF, VEGFR, and PDGFR, acting primarily as an antineoplastic through dual antiproliferative and anti-angiogenic effects. In preclinical and early clinical studies, the combination showed enhanced anti-tumor effect by inducing necrotic cell death distinct from apoptosis, targeting cells with high pDAPK^S308^ expression, and appeared generally well tolerated at lower doses. The developer of MG005 is Metagone Biotech; sorafenib is developed and marketed by Bayer/Onyx (as Nexavar). The main studied indication is advanced or refractory solid tumors; the combination is in early clinical development.
