This is a **combination therapy** consisting of two investigational cancer vaccines (**MG1-MAGEA3** and **Ad-MAGEA3**), the immune checkpoint inhibitor **pembrolizumab** (a PD-1 antagonist), and the chemotherapeutic agent **cyclophosphamide**. MG1-MAGEA3 and Ad-MAGEA3 are viral vector-based immunotherapies designed to induce an immune response against tumors expressing the MAGEA3 antigen. Pembrolizumab blocks the PD-1 receptor on T cells, thereby enhancing anti-tumor immune responses. Cyclophosphamide is a cytotoxic alkylating agent, often used here at low (metronomic) doses, partly to deplete regulatory T cells and further stimulate anti-tumor immunity. This combination aims to induce robust, antigen-specific tumor immunity and synergize with immune checkpoint inhibition for enhanced anti-cancer efficacy.
