MGA012 + MGD007 is a combination therapy being investigated for the treatment of relapsed or refractory metastatic colorectal carcinoma. This combination pairs two distinct therapeutic agents with complementary mechanisms of action. MGA012 (also known as INCMGA00012 or retifanlimab) is an anti-PD-1 antibody that works by blocking the PD-1 receptor, thereby enhancing T-cell immune responses against cancer cells. MGD007 is a humanized gpA33 × CD3 DART® (Dual-Affinity Re-Targeting) protein developed by MacroGenics. This bispecific T-cell engager works by simultaneously binding to: gpA33 (a surface antigen expressed on colorectal cancer cells) and CD3 (a T-cell surface glycoprotein). This dual binding mechanism redirects T cells to target and kill cancer cells expressing the gpA33 antigen.
