MGD013 (tebotelimab) + brivanib alaninate is an investigational combination therapy studied primarily for advanced liver cancers, including hepatocellular carcinoma (HCC). MGD013, also known as tebotelimab, is a bispecific DART® monoclonal antibody that targets and blocks both PD-1 and LAG-3 immune checkpoint molecules to restore or sustain T-cell function in cancer immunotherapy. Brivanib alaninate (BMS-582664) is an oral small molecule multitargeted tyrosine kinase inhibitor that antagonizes VEGFR2 and FGFR1, key pathways involved in tumor angiogenesis and growth. The combination aims to enhance anti-tumor activity by simultaneously modulating immune checkpoints and inhibiting tumor angiogenesis. The drugs have been evaluated together in phase 1/2 clinical trials for advanced HCC, including patients who have failed prior immune checkpoint inhibitor therapy[1][4][6][8].
