Overview

microRNA-424 + microRNA-503 is a microRNA-based therapeutic combination consisting of two miRNAs that are part of a conserved cluster. In the context of oncology, particularly ovarian cancer, these miRNAs function as tumor suppressors by directly targeting and inhibiting the expression of the WEE1 G2 checkpoint kinase. Their downregulation is associated with the maintenance of cancer stem-like cell (CSC) properties and resistance to platinum-based chemotherapy. Experimental restoration or overexpression of the miR-424/503 cluster has been shown to suppress CSC activity, re-sensitize chemoresistant ovarian cancer cells to carboplatin, and reduce tumor growth and peritoneal seeding in preclinical models.
