Micvotabart pelidotin + pembrolizumab is a combination therapy being developed by Pyxis Oncology in collaboration with Merck. Micvotabart pelidotin (formerly PYX-201) is a first-in-concept antibody-drug conjugate (ADC) that targets the extradomain-B of fibronectin (EDB+FN), a non-cellular structural component of the tumor extracellular matrix that is selectively overexpressed in various solid tumors. The ADC is designed to exert anti-tumor activity through direct cell killing, bystander effects, and the induction of immunogenic cell death. Pembrolizumab is a monoclonal antibody that inhibits the programmed cell death protein 1 (PD-1) receptor, thereby enhancing the immune system's ability to detect and destroy cancer cells. This combination is currently being evaluated in Phase 1 clinical trials for the treatment of recurrent/metastatic head and neck squamous cell carcinoma (R/M HNSCC) and other solid tumors.
