Overview
Midazolam is a short‐acting benzodiazepine used for anesthesia, procedural sedation, trouble sleeping, and severe agitation. It acts as a positive allosteric modulator of the GABA-A receptor, producing sedative, anxiolytic, muscle relaxant, anticonvulsant, and amnesic effects. Toreforant (JNJ‑38518168) is an oral small molecule antagonist of the histamine H4 receptor developed by Janssen/Johnson & Johnson for immune-mediated diseases such as rheumatoid arthritis and asthma. Toreforant also modulates Th1/Th2/Th17 cell responses. The combination of these two drugs has been studied in clinical trials primarily to assess pharmacokinetic interactions rather than for a specific therapeutic indication[1][3][6][8].
