Drug intelligence / Profile preview
**Milademetan + atezolizumab** is an investigational combination of two distinct anticancer drugs: milademetan, a small molecule oral inhibitor of the MDM2-p53 complex that reactivates p53 function, and atezolizumab, a monoclonal antibody immune checkpoint inhibitor targeting programmed death-ligand 1 (PD-L1). This combination is being evaluated primarily in advanced solid tumors with CDKN2A loss and wildtype p53, taking advantage of milademetan’s MDM2 inhibition to sensitize tumor cells and atezolizumab’s ability to enhance immune-mediated tumor cell killing. Both drugs are being studied together to offer a potentially synergistic approach in genetically defined patient populations[1][3][9].
