Miltuximab F(ab')2 is a bivalent antibody fragment derived from the monoclonal antibody miltuximab (also known as MIL-38), which targets glypican-1 (GPC-1). Developed by GlyTherix, the F(ab')2 fragment is engineered to lack the Fc domain, which facilitates faster systemic clearance and potentially improved tumor penetration compared to the full-length IgG antibody. It is primarily being evaluated as a targeting moiety for radiopharmaceuticals, such as zirconium-89 (89Zr) labeled immuno-PET tracers, for the non-invasive detection and monitoring of GPC-1-overexpressing tumors. Preclinical studies in glioblastoma models have investigated its utility in molecular imaging to identify GPC-1-positive disease, offering a different pharmacokinetic profile than the whole antibody format.
