Drug intelligence / Profile preview
ZT-1 (mimopezil) is a small molecule prodrug of huperzine A, a reversible and selective acetylcholinesterase (AChE) inhibitor originally derived from the Chinese herb *Huperzia serrata*. Developed by Debiopharm, ZT-1 is a Schiff base derivative designed to improve the pharmacokinetic profile and safety of huperzine A for the treatment of Alzheimer's disease. Upon administration, ZT-1 is rapidly absorbed and converted into its active metabolite, huperzine A, which increases acetylcholine levels in the brain by inhibiting its degradation. Clinical development has explored both oral formulations and a monthly subcutaneous sustained-release implant (the BRAINz study), the latter intended to provide continuous drug delivery, improve patient compliance, and reduce the variability in drug exposure compared to daily oral AChE inhibitors.
