Overview

Mini CYVE is an attenuated, multi-agent chemotherapy regimen primarily used in the treatment of pediatric mature B-cell lymphomas, including Burkitt lymphoma, diffuse large B-cell lymphoma (DLBCL), and B-cell acute lymphoblastic leukemia (B-ALL). It is a reduced-intensity version of the standard CYVE consolidation regimen from the LMB-89 protocol, specifically designed to minimize long-term toxicities such as infertility (from high-dose cyclophosphamide) and cardiotoxicity without compromising survival outcomes. The regimen consists of cytarabine (ara-C), etoposide, and dexamethasone administered at attenuated doses. It was evaluated in the FAB LMB96 international randomized factorial trial led by Gustave Roussy and the French-American-British (FAB) consortium. The mechanism of action involves DNA synthesis inhibition by cytarabine and topoisomerase II inhibition by etoposide, supported by the anti-inflammatory and immunosuppressive effects of dexamethasone.
