Overview

Minocycline + edetic acid (M-EDTA) is a combination catheter lock solution developed by the University of São Paulo and the Integrated Centre of Nephrology for use in hemodialysis patients with stage 5D chronic kidney disease. The solution combines the tetracycline antibiotic minocycline (3 mg/mL) with the chelating and anticoagulant agent edetic acid (EDTA, 30 mg/mL). Minocycline provides broad-spectrum antibacterial activity by inhibiting the 30S ribosomal subunit, while EDTA acts as an anticoagulant and a sensitizing agent that disrupts bacterial biofilms by chelating divalent cations like calcium and magnesium. This dual action is intended to prevent catheter-related bloodstream infections (CRBSI) and maintain catheter patency by reducing the incidence of poor blood flow rates, as evaluated in the Phase 4 CLOCK trial.
