Overview

Minocycline + methotrexate is a combination of two small molecule drugs used primarily in the treatment of rheumatoid arthritis. Minocycline is a tetracycline-class antibiotic with anti-inflammatory properties, while methotrexate is an antimetabolite and antifolate agent that acts as a disease-modifying antirheumatic drug (DMARD). The combination has been studied for its potential to improve symptoms and slow disease progression in early rheumatoid arthritis. Minocycline exerts its effects by inhibiting bacterial protein synthesis and modulating inflammatory pathways, whereas methotrexate inhibits dihydrofolate reductase, interfering with DNA synthesis and immune cell proliferation. Clinical studies suggest the combination may be well tolerated but does not significantly increase efficacy or toxicity compared to monotherapy[3][4][6].
