Overview

Minocycline + metronidazole is a combination of two small-molecule antibiotics used primarily for the treatment of infections where both anaerobic and aerobic bacteria are implicated. Minocycline is a semisynthetic tetracycline antibiotic that inhibits bacterial protein synthesis by binding to the 30S ribosomal subunit, thereby preventing the addition of amino acids to growing peptide chains. Metronidazole is a nitroimidazole antibiotic that exerts its effect by disrupting DNA synthesis in anaerobic bacteria and certain protozoa, leading to cell death. This combination has been studied for enhanced efficacy in treating periodontal diseases (such as periodontitis and peri-implantitis) and macrolide-resistant Mycoplasma genitalium infections, showing synergistic antibacterial effects, reduction in inflammatory markers (e.g., IL-1β, TNF-α), improved clinical outcomes, and reduced infection rates compared to monotherapy[1][2][3][4][5][8]. The regimen may be administered locally (as ointment/gel) or orally depending on indication.
