Minocycline + N-acetyl cysteine is an investigational combination therapy consisting of minocycline, a tetracycline-class antibiotic with anti-inflammatory properties, and N-acetyl cysteine (NAC), an antioxidant amino acid derivative. This combination has been studied primarily in preclinical models for its synergistic neuroprotective effects following traumatic brain injury (TBI) and retinal ischemia. The mechanism involves modulation of inflammation, reduction of oxidative stress, preservation of myelin integrity, and improvement in cognitive outcomes post-injury. Minocycline acts by inhibiting microglial activation and pro-inflammatory cytokines, while NAC replenishes intracellular glutathione levels to counteract oxidative damage. Both agents are FDA-approved individually for other indications but not as a fixed-dose combination[1][3][5].
