Overview

This combination therapy consists of miransertib (ARQ 092), an oral, potent, selective, and allosteric pan-AKT inhibitor, administered alongside the standard chemotherapy agents carboplatin and paclitaxel. Miransertib targets AKT1, AKT2, and AKT3, inhibiting the PI3K/AKT/mTOR pathway, which is frequently dysregulated in various cancers. Carboplatin acts as a DNA alkylating agent, while paclitaxel stabilizes microtubules to inhibit cell division. Developed by ArQule (a Merck subsidiary), this combination was evaluated in Phase 1b clinical trials for advanced solid tumors, specifically focusing on ovarian and endometrial cancers, particularly those harboring PIK3CA, PIK3R1, or AKT1 mutations.
