This refers to the chemotherapy regimen utilized in the Phase 2 CHART-AC clinical trial (NCT07425054), led by Dr. Jennifer Dorth for the treatment of previously untreated locally-advanced anal cancer. The regimen consists of mitomycin-C (12 mg/m²) administered on day 1, combined with a fluoropyrimidine backbone of either capecitabine (825 mg/m² twice daily on all days of radiotherapy) or 5-fluorouracil (1000 mg/m²/day as a continuous infusion for 96 hours on days 1-4 and 29-32). In the context of the CHART-AC trial, this chemotherapy is administered concurrently with radiation therapy (chemoradiation), with the total doses of both radiation and chemotherapy being customized based on the patient's human papillomavirus (HPV) circulating tumor DNA (ctDNA) response during treatment. Patients with unfavorable ctDNA responses may also receive the PD-1 inhibitor retifanlimab following the completion of chemoradiation.
