Overview

This is a combination chemotherapy regimen consisting of three agents: - **Mitomycin** (also known as mitomycin-C or MMC), an antitumor antibiotic that acts as an alkylating agent causing DNA cross-linking and inhibition of DNA synthesis. - **Doxifluridine** (5'-deoxy-5-fluorouridine or 5'-DFUR), an oral prodrug of 5-fluorouracil (5-FU) that is converted in the body to active 5-FU, which inhibits thymidylate synthase and disrupts DNA synthesis. - **Cisplatin** (CDDP), a platinum-based chemotherapeutic agent that forms DNA cross-links and induces apoptosis. The combination has been studied primarily for the treatment of advanced unresectable or resected gastric cancer. The rationale for combining these drugs includes potential synergistic effects between mitomycin and both cisplatin and fluoropyrimidines. Clinical trials have evaluated this regimen in adjuvant settings after gastrectomy as well as in advanced disease[1][2][3][4]. The addition of cisplatin to mitomycin plus doxifluridine was found to be safe but did not significantly improve survival outcomes compared to regimens without cisplatin[2][4].
