Mitoxantrone + DaunoXome is a combination chemotherapy regimen consisting of mitoxantrone, a synthetic antitumor antibiotic and DNA intercalator, and DaunoXome, a liposomal formulation of daunorubicin. Mitoxantrone disrupts DNA synthesis and repair by intercalating into DNA and inhibiting topoisomerase II, leading to cytotoxicity in rapidly dividing cells[3][8]. DaunoXome delivers daunorubicin encapsulated in liposomes to enhance tumor selectivity; daunorubicin also acts as an anthracycline antibiotic that intercalates into DNA and inhibits topoisomerase II activity[6][7]. This combination may be used experimentally or off-label for the treatment of certain leukemias or refractory cancers where both agents' mechanisms are leveraged for synergistic cytotoxic effects.
