This is a combination chemotherapy regimen consisting of three agents: mitoxantrone, etoposide, and gemtuzumab ozogamicin. Mitoxantrone is an anthracenedione antineoplastic agent that intercalates into DNA and inhibits topoisomerase II, leading to DNA strand breaks and inhibition of cell replication. Etoposide is a topoisomerase II inhibitor that causes DNA damage by preventing the re-ligation of DNA strands during replication. Gemtuzumab ozogamicin is an antibody-drug conjugate composed of a humanized monoclonal antibody targeting CD33 linked to the cytotoxic agent calicheamicin; upon binding to CD33-positive cells (such as those in acute myeloid leukemia), it delivers calicheamicin intracellularly, causing double-stranded DNA breaks and apoptosis[1][2][3][4][5][6]. This combination has been studied primarily as salvage therapy for patients with acute myeloid leukemia (AML) who have failed initial induction therapy[4][5].
