Drug intelligence / Profile preview
Mitoxantrone hydrochloride liposome (PLM60) is a liposomal formulation of the synthetic anthracenedione antineoplastic agent mitoxantrone. Developed by CSPC Pharmaceutical Group, it is designed to improve the pharmacokinetic profile of conventional mitoxantrone by increasing circulation time and enhancing accumulation in tumor tissues while reducing systemic toxicities, particularly cardiotoxicity. Mitoxantrone acts as a DNA intercalator and a topoisomerase II inhibitor, leading to DNA strand breaks and inhibition of DNA synthesis. In the context of the First Affiliated Hospital Xi'an Jiaotong University, it is being evaluated as a component of a modified BEAM (or BEAC) conditioning regimen (including carmustine, etoposide, and cytarabine) for patients with non-Hodgkin's lymphoma undergoing autologous hematopoietic stem cell transplantation (ASCT). The liposomal formulation was approved by China's NMPA in 2022 for the treatment of relapsed or refractory peripheral T-cell lymphoma.
