MK-0893 + sitagliptin is an investigational combination of two oral small molecule drugs for type 2 diabetes. **MK-0893** is a glucagon receptor antagonist that works by blocking the glucagon receptor, thereby reducing hepatic glucose output. **Sitagliptin** is a dipeptidyl peptidase-4 (DPP-4) inhibitor that increases endogenous incretin levels (GLP-1, GIP), promoting insulin release and decreasing glucagon secretion in response to food. The combination is designed to act synergistically to improve glycemic control by reducing both hepatic glucose production and increasing insulin-dependent glucose clearance.
