MK-6213 + atorvastatin is an experimental oral combination therapy investigated for the treatment of hypercholesterolemia and related lipid disorders. MK-6213 is a small molecule inhibitor of Niemann-Pick C1-like protein 1 (NPC1L1), acting as a cholesterol absorption inhibitor, developed by Merck Sharp & Dohme. Atorvastatin is a 3-hydroxy-3-methylglutaryl-coenzyme A (HMG-CoA) reductase inhibitor ("statin"), widely used for lowering cholesterol by inhibiting endogenous cholesterol synthesis. The combination was tested to achieve more substantial lipid-lowering effects than monotherapy, leveraging complementary mechanisms: inhibition of intestinal cholesterol absorption (MK-6213) and inhibition of hepatic cholesterol synthesis (atorvastatin).
