MK-6598 + pembrolizumab is an investigational combination therapy consisting of MK-6598, an orally administered small molecule inhibitor of Interleukin-4-Induced Gene 1 (IL4I1), and pembrolizumab, a monoclonal antibody targeting the programmed cell death protein 1 (PD-1) receptor. MK-6598 aims to reverse tumor-mediated immune suppression by blocking IL4I1, thereby reducing hydrogen peroxide production in the tumor microenvironment and enhancing anti-tumor immune response. Pembrolizumab blocks PD-1, releasing the brake on T-cells and thereby potentiating immune-mediated tumor cell killing. This combination seeks to augment immunotherapy efficacy by addressing complementary mechanisms of immune escape. It is being studied for the treatment of advanced or metastatic solid tumors.[1][2][3]
