Overview

MK-7162 + pembrolizumab is an investigational combination therapy developed by Merck (MSD) for the treatment of advanced solid tumors. The combination pairs MK-7162, a potent and selective small-molecule antagonist of the adenosine A2A receptor (A2AR), with pembrolizumab (Keytruda), a humanized monoclonal antibody that inhibits the programmed cell death protein 1 (PD-1) receptor. Adenosine accumulation in the tumor microenvironment is a known mechanism of immune evasion, as it suppresses T-cell and NK-cell activity via A2AR signaling. By blocking A2AR, MK-7162 is intended to reverse this immunosuppression and work synergistically with pembrolizumab's checkpoint inhibition to enhance anti-tumor immune responses. The combination has been evaluated in Phase 1 clinical trials, including NCT03364049, to determine safety, tolerability, and preliminary efficacy in patients with various advanced or metastatic malignancies.
