MK-8033 is a potent small-molecule inhibitor of the hepatocyte growth factor (HGF)/c-Met axis. It binds preferentially to the activated conformation of c-Met and has demonstrated anti-tumor activity in preclinical models, particularly in gastric and non-small cell lung cancer cell lines. Inhibition of c-Met by MK-8033 leads to reduced autophosphorylation and downstream signaling through ERK1/2 and AKT pathways. Omeprazole is a proton pump inhibitor (PPI) that suppresses gastric acid secretion by inhibiting the H+/K+ ATPase enzyme system at the secretory surface of gastric parietal cells. The combination was studied in clinical trials to assess pharmacokinetic interactions; omeprazole did not affect MK-8033 absorption. Development of MK-8033 was discontinued after phase I studies due to limited clinical activity[2][6][7].
