Overview

MK-8189 + atypical antipsychotic is a combination treatment regimen consisting of MK-8189, a selective phosphodiesterase 10A (PDE10A) inhibitor, and an atypical antipsychotic agent. MK-8189 increases intracellular cyclic nucleotide levels in medium spiny neurons in the striatum, affecting dopamine signaling by mimicking the net effects of dopamine D1 receptor agonists and D2 receptor antagonists. Atypical antipsychotics, including drugs such as risperidone, target multiple receptors (notably dopamine D2 and serotonin 5-HT2A receptors) and are standard first-line treatments for schizophrenia. The combination is under exploration to assess whether MK-8189, which may impact both positive and cognitive symptoms of schizophrenia with possibly fewer metabolic side effects, can provide additive or synergistic benefits when used alongside established atypical antipsychotics[1][3][5].
