MK-8353 + pembrolizumab is an investigational combination therapy for advanced solid tumors. **MK-8353** is an orally bioavailable, dual-mechanism, highly selective small molecule inhibitor of extracellular signal-regulated kinases 1 and 2 (ERK1/2), components of the MAP kinase pathway, developed for its activity against cancers with MAPK pathway alterations, such as BRAF or RAS mutations. **Pembrolizumab** is a monoclonal antibody that binds programmed death-1 (**PD-1**) receptor on immune cells, blocking its interaction with ligands PD-L1/PD-L2 and thus enhancing T-cell–mediated immune responses against tumors. This combination is studied to evaluate whether dual ERK inhibition and immune checkpoint blockade may have synergistic antitumor effects, particularly in heavily pretreated patients with advanced solid tumors or microsatellite stable colorectal cancer[1][2][3][5].
