Overview

MM-111 + paclitaxel + trastuzumab is an investigational combination regimen for the treatment of HER2-positive cancers, particularly gastroesophageal (GE) and gastric adenocarcinomas. MM-111 is a bispecific antibody that targets both HER2 and HER3, forming an inhibitory complex that blocks HER3 and downstream PI3K pathway activation in HER2-overexpressing cancers. Paclitaxel is a microtubule-stabilizing cytotoxic chemotherapeutic. Trastuzumab is a monoclonal antibody that targets the HER2 receptor, inhibiting HER2-mediated signal transduction and inducing antibody-dependent cellular cytotoxicity. Combining these agents aims to enhance antitumor activity by simultaneously inhibiting HER2/HER3 signaling and enhancing cytotoxic effects. The regimen was evaluated in phase I/II trials in HER2-positive GE cancers, but phase II trials were terminated early due to adverse outcomes in overall survival and progression-free survival when MM-111 was added to paclitaxel and trastuzumab[1][3][4].
