Overview

MM-151 + irinotecan is a combination therapy under investigation for the treatment of metastatic colorectal cancer, particularly in patients with RAS wild-type tumors. MM-151 is an oligoclonal mixture of three fully human monoclonal antibodies targeting non-overlapping epitopes on the epidermal growth factor receptor (EGFR), designed to overcome resistance mechanisms seen with other anti-EGFR therapies. Irinotecan, including its liposomal formulation ONIVYDE (nal-IRI or MM‑398), is a topoisomerase I inhibitor used in chemotherapy regimens for colorectal and pancreatic cancers. The combination aims to block EGFR-driven oncogenic signaling and inhibit DNA replication in tumor cells, providing a dual mechanism against tumor growth and survival[1][2][5][6][7].
