MM-151 + trametinib is a combination investigational therapy of MM-151, an anti-EGFR monoclonal antibody, and trametinib, a selective MEK1/2 inhibitor. MM-151 targets and blocks multiple epitopes of the epidermal growth factor receptor (EGFR), inhibiting ligand-induced activation and downstream cancer-promoting signaling. Trametinib inhibits MEK1 and MEK2 enzymes in the MAPK/ERK pathway, preventing cancer cell proliferation driven by RAS-RAF-MEK-ERK signaling. This combination aims to overcome resistance mechanisms in tumors, particularly those reliant on EGFR and MAPK pathway activation, including certain colorectal cancers and other advanced solid tumors.
