MM-302 + trastuzumab + cyclophosphamide is a combination regimen investigated for the treatment of advanced or metastatic HER2-positive breast cancer. MM-302 is a HER2-targeted PEGylated antibody–liposomal doxorubicin conjugate that selectively delivers the cytotoxic agent doxorubicin to HER2-overexpressing tumor cells, thereby limiting exposure to healthy tissues such as cardiomyocytes. Trastuzumab is a monoclonal antibody targeting the extracellular domain of the human epidermal growth factor receptor 2 (HER2), inhibiting downstream signaling and mediating immune responses against tumor cells. Cyclophosphamide is an alkylating agent that interferes with DNA replication and cell division. Preclinical studies demonstrated enhanced antitumor activity when these agents were combined, particularly in anthracycline-naïve models. Clinical trials showed that this combination was generally well tolerated and exhibited promising efficacy in patients with advanced HER2-positive breast cancer; however, subsequent phase II trials did not demonstrate improved progression-free survival compared to standard regimens[1][2][3][5]. MM-302 was developed by Merrimack Pharmaceuticals.
