MNKD-101 (MannKind)

MNKD-101 is an investigational inhaled formulation of benralizumab, a humanized monoclonal antibody, being developed by MannKind Corporation for the treatment of eosinophilic asthma. Utilizing MannKind's proprietary dry powder inhalation technology, MNKD-101 is designed to deliver the therapeutic agent directly to the lungs, potentially enhancing local efficacy while minimizing systemic exposure. Benralizumab targets the alpha subunit of the interleukin-5 receptor (IL-5Rα) expressed on the surface of eosinophils and basophils. By binding to IL-5Rα, the antibody prevents the binding of interleukin-5 (IL-5) and triggers antibody-dependent cell-mediated cytotoxicity (ADCC), primarily through natural killer (NK) cells, leading to the rapid and near-complete depletion of eosinophils in the airways.
